Preparation of Culture Media

The asymbiotic method of seed culture involves germination seeds and growing the seedlings upto the transplanting stage under sterile conditions in an agar medium which is supplemented by all the necessary nutrients required by the plant . The nutrient solution will have the following basic components :

1. Inorganic salts

2.Carbohydrates

3. Organic substances like vitamins , amino acids and growth hormones.

Several standardised media are widely in use now. The earliest to be formulated and still widely used , is the Knudson 'C' medium , the composition of which is given below :

Calcium nitrate 1.00 gm
Monobasic Potassium Phosphate 0.25 gm
Magnesium sulphate o. 25 gm
Ammonium sulphate 0. 50 gm
Sucrose 20.00 gm
Ferrous sulphate 0.025 gm
Manganese sulphate 0. 0075 gm
Agar 15 gm
